Scientists have known that infection inhibits tumour growth for over 100 years. Until now, they assumed that an immune system highly mobilised to deal with one threat responds better to another, and that this explained the finding. But the new research suggests that the immune system has nothing to do with it. According to Hunter of the University of Pennsylvania and his colleagues, infection stimulates the production of molecules that prevent angiogenesis, or the growth of new vascular tissue, which forms the tumour's life support system. Laham of the Angiogenesis Research Center at Harvard Medical School in Cambridge, Massachusetts, says the study provides the first direct evidence that infection itself is important, without an immune reaction. " This has all of us worried as well as elated, " he says. " Worried because it means there is much more to learn about the infectious process, and we are all exposed to infection all the time. Elated because infectious agents are easier to control than genetic predispositions to cancer. " Mystery culprit The researchers implanted malignant melanomas in mice genetically engineered to lack immune cells such as T-lymphocytes or macrophages. They found that even in the absence of the normal immune defences those tumours stopped growing when the mice were infected - in this case, with the protozoan parasite Toxoplasma gondii. Even when the team tried to artificially stimulate angiogenesis in T. gondii-infected mice by injecting potent angiogenic compounds such as basic fibroblast growth factor (bFGF), they saw no new growth of blood vessels. The identity of the anti-angiogenic molecules produced in response to infection remains a mystery. But by comparing the blood of the infected animals with those of healthy animals the researchers hope to be able to isolate them. " We are very much interested in identification of these 'culprits' because we believe that they will be extremely useful therapeutic agents not only for cancer but also for other diseases where unwanted angiogenesis takes place, " says one of the researchers, Andrei -Tikhonenko. One such disease is endometriosis, which affects the lining of the womb and is strongly associated with female infertility. Several anti-angiogenic compounds have been identified to date, but they have yet to be proven effective in clinical trials.
