I will most likely be travelling either in Canada or into the US. I really want to find someone who can take care of me en bloc! The consult I had yesterday scared me a bit.. Dr. Brown told me that he won't remove the capsules because of the risk of trauma, bleeding and even having a lung punctured because my implants are under the muscle:( This really scared me... I had my chest wall detatched 7 times total............no lung punctured, you still have facia and bones ( ribs) there ............he is just not skilled enough to do yours and it is good he told you, instead of doin it and ruining you for life. Our targeted proteomics approach was aimed at identification of the proteins adsorbed to the surface of silicone because they have been characterized as key components in the onset and perpetuation of local immune reactions to silicone. The composition of the proteinacious film, the dynamics of protein deposition, and protein modifications after adsorption were analyzed both in vivo and in vitro. Differential analysis of protein deposition was performed, followed by protein identification with mass spectrometry, database matching, and Western blots. Thus far, we have identified the 30 most abundant proteins deposited on the surface of silicone, the largest known inventory of such proteins so far. Structural and extracellular matrix proteins predominated, followed by mediators of host defense, metabolism, transport, and stress related proteins. In addition, several biochemical modifications of fibronectin, vitronectin, and heat shock protein 60 were detected. Our analyses also revealed previously undetected proteins deposited on the surface of silicone. As tentative initiators and/or modulators of the response to silicone, they are therefore valuable candidates for prognosis and therapy. The objective of these studies was to determine which autoimmune-like symptoms and other symptoms are associated with implanted materials. OST has developed a rat model to study and understand immune responses to silicone gel and oil used in breast implants. This model included normal rats and a strain of autoimmune rats, that mimicked clinical conditions found in patients with silicone gel breast implants. Mixtures of silicone gel/oil were injected into the mammary area of female rats. Scientists then measured the levels of autoantibodies to collagen and to nuclear proteins developed over 2 years in response to these mixtures. Results from the rat model indicate that specific autoantibodies may be induced by certain biomaterials. The immune system recognizes a biomaterial-connective tissue protein association as altered-self or as foreign. OST scientists have demonstrated that medical grade silicone oil can stimulate serum autoantibodies against collagen and against DNA when this oil is injected into mammary tissues of rats. Autoantibody production against connective tissue proteins is an immune response that is consistent with reports from women with silicone breast implants. The results also demonstrated pathological changes in animals that may result from the autoimmune response and that silicone gel can migrate to distant anatomical sites or localize at the implantation site. This work has been presented at The FDA Science Forum, The American Association of Immunology, and The American Association of Biochemistry and Molecular Biology. The implication from these findings is that leaked oil from a breast implant via leaching or with rupture might provide stimulus for the production of autoantibodies in clinical patients. Therefore, OST scientists evaluated autoantibody levels (in blinded experiments) in serum samples from 150 patients representing four groups: women with silicone implants without connective tissue disease, women with silicone implants with connective tissue disease (CTD), women with connective disease but no implants, and healthy women volunteers. Results from these experiments show in a statistically significant manner that elevated autoantibodies to collagen type I, collagen type II and anti-DNA were detected in serum of patients with CTD, CTD + silicone implants, and silicone implants without CTD. Using two different assays, autoantibodies to connective tissue proteins (e.g., collagen) and to DNA and intranuclear proteins have been detected in women with silicone breast implants. Historically, there is a strong correlation between anti-nuclear antibodies and clinical symptoms of some autoimmune diseases. OST has documented serum immune responses in these patients with the goal of enhancing the ability to predict the likelihood of immunotoxic symptoms occurring in the presence of implants, including breast prostheses. This work has been presented to the Institute of Medicine. OST scientists are continuing to investigate these results by correlating this data to the clinical history of the patients. Researchers plan to determine the clinical significance of the data and to study a larger patient population well defined with regard to implants and autoimmune disease symptoms. Thank you for being such a trooper and such a researcher! You help me everyday with your knowledge and your strength... I was really scared about removing the capsules after that last appointment, but know because of all the women here that it is what must be done in order for me to recover from these implants. I have a few more surgeons to talk with before I make my final decision, but the good news is that I have managed to find a way to borrow most of the money, so if I can save some too, I should be able to get my explant in August even if I end up travelling to Denver! Guest guest Posted April 27, 2007 Report Share Posted April 27, 2007 Katy ~ Yep, 7 ! ! amazing. huh? my body just kept attacking the free floating silicone and growing scar tissue around it and cysts and fat necrosis, and all kinds of nasties. Very painful and would cause burning and numbness in my chest and arms and up my neck. It was a total nightmare.......One got so big it filled an entire urine cup. Took her 6 hours to get it out, it even went up to my collarbone and wrapped around the bone and intertwined with an artery. That is the one that did not show up on the 5 xrays and 2 mri's.....Gross ! ! My capsule blew out due to the mammogram, so the silicone had free range in my chest wall for 2 weeks. Many ruptures are still contained in the scar capsule. I dont recommend a mammogram if a rupture is suspect, could burst the scar capsule, it did mine ! ! Hugs ~ DedeSee what's free at AOL.com.
