I'm not a pharmicist but in my experience there are two elements of concern.

First if the shot aren't refridgerated MB12 can break down over time, but it

would take

more than 5 days for it to be significant.

Second and most importantly is if they become contaminated. I would ask the

pharmacy's

opinion. If I had to go through that much trouble and I was really wanting to

try it... at 5

days... I would be torn about it because I have not had shots out that long. If

I was in the

states I wouldn't use them. To play it safe I wouldn't use them.

Using the spray when it's out is typically not a problem because it doesn't have

to be

perfectly sterile... but as an injectable your looking for trouble. Basically

you're concerned

about an infection.

I probably wouldn't do it (you can tell I'm thinking about it as I write). If I

was going to do

it I would inject myself first and see if it stung on the way in or within 24

hours.

Basically, I probably wouldn't consider it without the pharmacy telling me (or

parents

telling me) that they have done it successfully.

Italian pharmacy we use has begun shipping the MB12 in a white styrofoam

insulated box along with ice packs, especially now that the hot weather

has arrived. We have ours delivered overnight within Italy but I don't

know what the normal delivery time is for outside-Italy shipments in the

EU. Earlier this year they tried shipping my order via a service that

was to have delivered it to my door instead of me going to the courier

to pick it up - 9 days later still no package and on the 10th day, I

refused the package because it had been unrefrigerated too long...the

pharmacy replaced it with overnight shipping. The shots do not

contain preservatives which is why it is critical that they be

refrigerated. Again, call the pharmacy - the two I know who handle the

MB12 are very customer-friendly...and they take great care to ensure

product safety.

contact the pharmacy and ask them about this, as well as consider the

possibility that this batch is a dud.

Hi,

Could you provide me with the address or mail or phone of this pharmacy? My DAN also prescribed the shots & I live in Belgium, I’m afraid it’s might be difficult to get it from the states with this refrigeration problem. Altough service is much quicker from the states than inside Europe in general.

HI;

I LIVE IN THE UK (NOT A HUGE CITY) AND GET MY STUFF

FROM COASTAL COMPOUNDING IN THE STATES, IT IS VERY

WELL PACKED AND GETS HERE IN ABOUT 3 DAYS...

the only thing is you need to be home when the package

is delivered because you have to pay duty before they

will release it, otherwise it goes back to their

warehouse-

Hi Kendra,

Could you please tell me how you ordered your MB12 shots

internationally from Coastal? When I called, they specifically

required an US Rx. I live in Canada and they do not accept Canadian

Rx. Thanks.

Hey Stan and lsitmates,

Thanks to Guillaume, who alerted me to the posts, here is a little

bit of background on refrigeration issues with regard to MB12 shots.

Oct 23, 2005 history from Dr. Neubrander on refrigeration of MB12

shots -- When they first started compounding with Hopewell(May 2002

to February or March 2003), they were never refrigerated because PCAA

the distributor, recommend refrigeration for the base product, but

not the reconstituted product. Wwhen Coastal Compounding started also

compounding MB12 shots, they said to refrigerate them:

Oct 25, 2005 post to CK2 from Tyrus at Coastal Compounding - Coastal

Compounding chose to refrigerate based on good pharmacy practice.

Tyrus says here that while there is probably a very low chance of

contamination given the sterile conditions, good pharmacy compounding

practices are that injectibles made without a preservative should be

refrigerated until use (sterility issue):

Oct 23, 2005 post from Adam at Lee Silsby Compounding Pharmacy saying

that they have done independent stability testing and found that MB12

shots stored at room temperature for two weeks had a 30% reduction in

activity (lost their potency)

Hey All,

One more update on this for you all. The Italian pharmacy finally

got back to me. I asked specifically about it being in transit for 8

days. They replied that the shots can safely be stored for 8 days at

25-35 degrees centigrade (77-86 F). Lucky for us, it was nowhere

near that hot, as there were rain storms -- it was in the low 60s F

throughout that period.

Hi Theresa, just wanted to mention that we got a bottle of nasal mb12

from an Italian pharmacy (Legnani) but they did not ask for rx at that

time... This was a year ago, has the situation changed?

also, have you looked into or tried mb12 drops by Cenaverde, just heard

from someone who saw great results with those. no rx either.

Hey Natasa,

Thanks for this. I will check with them about the nasal spray. We

are waiting for our first consult with Dr. Mullan in a few weeks and

hope to get a prescription for the nasal spray.

Yes, we the started Cenaverde TD MB12 and it does seem to have an

effect. I didn't like some fo the ingredients in the transmedium

(part 2) so am using pure jojoba oil instead as a way to get it to

penetrate deeply.

I hope you are doing well! Have jsut been reading about calcium in

the mitochondria, at the same time that I'm reading about the effects

of B12 deificiency and elevated propionic acid in the mitochondria.

Since adding in a bunch of Krebs support, fatty acid metabolism

support (both a la Yasko) and much more B12 (Lulu has elevated

urinary MMA and FIGlu), Lulu has developped chicken skin. A friend

thinks that it may involve the export of calcium and the enhancement

of fatty acid metabolism -- that it's a good thing and that it will

pass as the excess calcium gets exported. I'm not sure that I'm

doing justice to the theory, but was wondering what your thoughts are

on B12 and calcium in the mitochondria?

We have studied several novel effects of vitamin B12 (cyanocobalamin)

on cellular Ca(2+) homeostasis in rat thymocytes. We determined the

effect of various concentrations of vitamin B12 on intracellular Ca

(2+) concentration ([Ca(2+)]i) and parameters of Ca(2+)in signaling

using the fluorescent dye Fura-2. The basal [Ca(2+)]i in Ca(2+)-

containing media was 115 +/- 5 nM but in vitamin B12 (10 nM)-treated

thymocytes [Ca(2+)]i was decreased to 60 +/- 15 nM (mean +/- SEM)

during the first 5 min. The decline in [Ca(2+)]i was accompanied by

an increase in the endoplasmic reticulum Ca(2+) store, presumably as

a result of Ca-ATPase activation. At the same time 100 nM-10 mM B12

induced the accumulation of Ca(2+) in mitochondria. Somewhat higher

concentrations of B12 (1-10 microM) had no effect on [Ca(2+)]i. A

further increase in B12 concentration with range from 50 microM to 1

mM caused a dose-dependent elevation of [Ca(2+)]i from the basal

level (115 +/- 5 nM) up to 200 +/- 50 nM in thymocytes, and this

elevation was partially blocked in Ca(2+)-free media. This high

concentration of vitamin B12 caused a gradual decrease of endoplasmic

reticulum Ca(2+) stores by means of Ca-ATPase inhibition. The B12-

induced increase in [Ca(2+)]i was not observed after depletion of

intracellular Ca(2+) stores, induced by addition of 2',5'-di(tert-

butyl)-1,4-benzohydroquinone (BHQ), an inhibitor of endoplasmic

reticulum Ca (2+)-ATPase, concanavalin A, or arachidonic acid. These

studies show that vitamin B12 regulates [Ca(2+)]i via several

different mechanisms at different B12 concentrations. Participation

of G proteins and calmodulin activity in B12-mediated [Ca(2+)]i
