Guest guest Posted September 22, 1998 Report Share Posted September 22, 1998 FYI Migraine Drug Eletriptan Absorbed Rapidly SEVILLE, SPAIN -- Sept. 22, 1998 -- Pfizer Inc.'s eletriptan, an antimigraine compound, is rapidly absorbed, exhibits predictable pharmacokinetics following oral dosing and appears to be well tolerated across age groups, according to a study reported at the third congress of the European Federation of Neurological Societies. Studies were conducted to evaluate the pharmacokinetics of eleptriptan in healthy male and females from age 18 to 93 and separately in subjects with renal and hepatic impairment. The study was presented by Milton, clinical project manager of Pfizer Ltd.'s central research facility in Sandwich, England. The study found that in healthy volunteers, systemic exposure is essentially linear over the dose range from 20 to 120 mg and that elimination is primarily by metabolism in the liver with only 10 percent of the dose excreted in urine. " Gender has no effect on the pharmacokinetics of eletriptan, " Dr. Milton said. " In the elderly some evidence was found of a small increase in exposure on eletriptan resulting in a longer half-life in this group. " We believe this is caused by a slower elimination rate and therefore decreased clearance in this age group. " In subjects with mild to moderate hepatic impairment, Dr. Milton reported some evidence of a small increase in exposure to eletriptan, probably related to decreased clearance and in those with mild, moderate or sever renal impairment the study found no significant effect on the pharmacokinetics of eletriptan.
