metformin, five hundred milligrams each, for ninety bucks. anybody found a

better price? also, a kilo of carnosine for five hundred bucks.

How do I find the site for these products?

[Metformin is a prescription drug but it can be purchased without a

In particular, Wakfer made a report on sci.life-extension

about a poster presentation at the Seventh International Symposium

on Neurobiology and Neuroendocrinology of Aging which we attended

in Bregenz Austria in July 2002. That study (which has never been

published) clearly showed that metformin at a human equivalent

dosage of about 1 gram per day extended the maximum lifespan of

mice almost as much as did calorie restriction. Many of the

physiological parameters were also similar. That poster and his

conversation with Roth (a senior author of the study) about

it were what decided him to restart metformin at 1 gram per day (for

both of us). Roth is no longer at NIA and is with his own company

which also is developing calorie restriction mimetics in competition

with Biomarker Research.

Those who decide to take metformin should, as usual, build up slowly

to their target dose by starting with a maximum of 1/4 of that

target dose and doubling every 2-3 weeks. Even after reaching target

with no untoward effects they should watch for symptoms of

hypoglycemia which is generally rare with this drug and for signs of

liver toxicity (by getting liver enzyme tests at least semi-

annually).

, I may have mistyped, I got 50, 500mg caps for $14.95, my

understanding was that you got 500, 500mg caps for $95.00. that would be a

saving of $55.00- shipping, so well worthwhile if I order more than one

product.

If I remember correctly there were studies showing positive correlation

between metformin intake and reduced testosterone levels in men. Any

new info on that subject? Right now I only take metformin as 'damage

control', a few minutes before I begin to eat a high carbohydrate

meal.

[i think so, yes. However I think caloric restriction also reduces levels of

testosterone. Metformin also increases homocysteine levels. Metformin is

hardly the holy grail for anti-aging, but I think it should be considered by

everyone over about age 40. It increases life-expentency in mice, as would be

expected given that it causes changes in gene expression that at least

partially mimic those from caloric restriction. Unfortunately no one seems to

have the data to show exactly how much metformin mimics the gene expression

from caloric restriction. The lastest issue of Life Extension Mag has a bar-

chart which on the surface appears to show that meformin mimics gene

expression 100%, but a closer look shows that the chart is visually

misleading: metformin was simply the most effective drug studied for mimicing

CR gene expression, so they set the bar for metformin at 100%. -gts]

I believe that TMG is supposed to hold down homocystien levels. Would one

[Yes, also folate, B-12 and B-6 help to control homocysteine. The same study

that showed an increase in homocysteine from metformin also showed reduced

levels of folate and B-12 (or B-6) from metformin so there is hope that

supplementing folate and B-12 and B-6 with metformin will keep metformin from

causing a rise in homocysteine. In any case it's probably a good idea for

metformin users to get their homocysteine checked periodically. -gts]

The link between the insulin receptor and longevity could help explain how

metformin extends life-expectency in lab animals and possibly also in

humans. Among other things, metformin appears to enhance sensitivity of

insulin receptors.

BOSTON -- August 15, 1997 -- Researchers at the Massachusetts General

Hospital (MGH) have discovered that a gene used by the tiny worm C. elegans

to regulate how much it eats, how fat it becomes and how long it lives is

strikingly similar to the gene for the human insulin receptor.

In today's issue of Science, the team led by Ruvkun, PhD, announced

their surprising discovery, implying that insulin-like control of metabolism

is very ancient. The finding also suggests that human counterparts of other

worm genes that interact with this insulin-like signal may be defective in

diabetes, an insight that could lead to new therapies for the disease. In

addition, the discovery that the worm's form of insulin regulates its

lifespan suggests that human aging may be intimately connected to how food

is metabolized.

Ruvkun and his colleagues-postdoctoral fellow Koutarou Kimura, PhD, graduate

student Heidi Tissenbaum, and technician Yanxia Liu-identified and cloned

the gene daf-2, which regulates whether C. elegans lives a short, lean life

focused on reproduction or a long, fat life focused on energy storage and

survival. They discovered that daf-2 has important structural similarities

to genes coding for insulin receptor proteins in humans and other mammals.
