Wednesday May 30 5:22 PM ET Cholesterol Drugs May Fight Inflammation, Too NEW YORK (Reuters Health) - If research in mice pans out, some cholesterol-lowering drugs may one day get makeovers that allow them to fight immune system-related diseases like rheumatoid arthritis.In experiments with several cholesterol drugs known as statins, investigators discovered how the medications help suppress inflammation in the body and showed that modified versions of statins can fight inflammatory disease in mice. They speculate that statin-like drugs may someday treat immune-related disorders marked by inflammation, and possibly help thwart other unwanted immune responses such as transplant rejection.Scientists at Novartis Pharma in Basel, Switzerland, and The Scripps Research Institute in La Jolla, California, report the findings in the June issue of Nature Medicine.The study was led by Dr. e Weitz-Schmidt of Novartis, a drugmaker that manufactures statin drugs.Statins are well known for their ability to lower unhealthy cholesterol levels, but research has also suggested the drugs may quiet inflammation set off by the immune system. For example, some studies of transplant recipients have suggested statins help suppress the immune response to foreign organs.In the new research, Weitz-Schmidt's team was able to uncover the possible reason statins have this ability. They found that several statins block the activity of an immune-system substance called lymphocyte function-associated antigen-1 (LFA-1). LFA-1 dwells on the surface of certain immune cells and helps orchestrate the body's inflammatory response.Moreover, when the scientists modified the statins to create an even stronger bond to LFA-1, the resulting compound suppressed abdominal inflammation in mice.These effects against inflammation are unrelated to the drugs' ability to lower cholesterol, the researchers note. Instead, by binding with it, certain statin compounds appear to block LFA-1.``This finding,'' the investigators write, ``indicates that statins might be able to interfere with anti-inflammatory processes and the immune response.''In the future, they speculate, compounds derived from statins might treat autoimmune diseases such as rheumatoid arthritis and psoriasis, or be useful in tissue transplants.SOURCE: Nature Medicine 2001;7:687-692.
