I don't want you to think I am starting an argument purely for the

sake of it. There are a lot of disagreements and confusions over the

role of serotonin on libido even in academia.

There is evidence that ssri's damage serotonin cells and leave you

with deplted serotonin neurons. I don't think this has been

considered very much partly because of an overgeneralisaztion that

serotonin is bad for the libido.

If you look at what each different serotonin receptor does, the

effects would be to cause very poor smooth muscle relaxation, and

poor sleep and wakefullness, as well as disrupting other systmes

indirectly.

5ht1 receptors can lower libiod, but in some parts of the brain they

may actually raise libido, which shows how complicated the matter is.

Yohimbine generally makes you feel very stressed, but some people

have found it usefull.

There are other examples of serotonin increasing libido within the

group. One member took a trycilic, long distance running seems to

work well (which increases serotonin and upregulates 5ht2a and

5ht2c). Also sleep deprivation, which has similar effects to long

distance running.

You say it does not really work reliably, so perhaps it is not

directly effecting the real problem. Sleep deprivation is probably

the best possible result, destroying every symptom of pssd completely.

Why does it work better combined with fluoxatine? 5ht2c receptors do

not inhibit libido, they increase it. Fluoxatine also contains a 5ht2

antagonist. Fluoxatine downregulates 5ht1a receptors, and also

increases serotonin quite a lot.

Naturalgal,

I noticed a positive change in libido the first couple of times I used

it. Then, probably due to the fact that nefazodone or its metabolytes

built up a little in my system, I lost the boost in libido after two

or three doses. By adding prozac to the mix, I prolonged the positive

effect of nefazodone.

As yet I have no standard regimen. I'm still experimenting with dosage

and frequency of use. It may take a while to pin it down. I do know

this: after trying moclobemide, bromocriptine, stablon, buspar, 5HTP,

vitamins etc. this seems the most promising so far.

I'm hoping I can get by with less than 50 mg of nefazodone, more like

30 mg a day at most, and 10 mg prozac every day or every other day. I

also want to add a small dose of yohmbine Hcl (5HT1 antagonist)to see

if it helps more.

Get a liver test to establish baseline levels of AST and ALT. I

noticed increase in libido with my very first 50mg dose which I took

at bedtime.

You're right that Buspar is a 5HT2 antagonist. I remembered

incorrectly. I should have said that Buspar is 5HT1 agonist.

As far as Nefazodone's antagonist action at 5HT2C goes, I really don't

know what to believe because its mCPP component also acts as an

agonist at 5HT2C

Buspar is mainly a 5ht1a agonist. I've read it might have some action

at other sites. After you've taken it for a few days the sleepiness

starts to wear off and you start to feel more awake.

Fluoxatine contains a 5ht2a/5ht2c antagonist, and also increases

dopamine and neuroadrenaline (unlike most other ssris).

The Nefazodone can metabolise into mCPP, but in its natural form it

does not contain a 5ht2c agonist, but purely contains a

5ht2c agonist. It also upregulates 5ht1a receptors indirectly, I have

read. I don't know which 5ht1a receptors it upregulates, but i have

read that some of them are important in libido, whilst some of them

reduce libido.

It is strange because first of all, 5ht2 receptors increase cGMP, so

if you block them you should have weaker erections. Do you think that

this stuff does improve nitric oxide or cGMP? because if it does,

maybe it is though an indirect pathway.

I think this discussion about receptrs is very important. It would be great to

know what's really going on, and you've covered some good ground. One thing

thoug - I think it would be more scientifically correct to say " it has 5HT2C

agonist activity " than " it contains a 5HT2C agonist " . It is after all, only one

molecule. The way you wrote it, it sounds like each medication " contains "

different things, but that's not the case. i hope what I just said doesn't

sound pedantic, but I think it's important to be clear with the terminology.

I would have thought Nefazodone porbably has mixed activity at 5ht2c

receptors. The drug itself is an antagonist at 5ht2 receptors, but it

has the potential to motabolise into mCCP, which is a very strong

agonist. Nutmeg is probably a 5ht2 agonist as well. The trycilic that

biker took also minly has activity at 5ht2c receptors (with regards

to its serotonin activity). It probably isn't as simple as just

having desensitized 5ht2c receptors I would have thought. But then

again, I would have expected people with pssd to have poor activity

at 5ht2c receptors; it seems to be an issue.

I would have expected that there might be a problem with a few other

5ht receptors, involving smooth muscle relaxation. Nitric oixde and

cGMP aren't just important for erections, they seem to be more

important than any other neuron for sleep and vigilance. From my

experiences with sleep deprivation I half expect that there is some

kind of complex signilling cascade either caused by poor sleep

quality or something else like poor cortisol or nitric oxide.

I 've been reading some posts by people who had positive results with

nefazodone and I thought that I 'd give it a try. Unfortunately, it

was pulled out of our market (Greece) because of the liver issues,

which are extremely rare. Does anybody have a link on where to order

it from (prescription free!)?
