I had taken 4 50 mg doses of Nefazodone about two weeks ago. I felt

groggy on the first half-tablet, but noticed, with each subsequent

dose, increased alertness accompanied by the usual libido decrease

and numbness that is typically associate with Prozac, Paxil, etc.

Discouraged, I stopped using the drug, believing it to be another

dead-end med. However, after a couple of days went by, I felt a

complete absence of numbness and a sure increase in libido, which has

continued undiminished for 10 days. Moreover, I felt a return of my

old self, more in the moment with less distracting thoughts racing

around in my head. In general, a very natural feeling of health.

At this point, I'm hoping that Nefazodone's antagonist action at 5HT2

persists after discontinuation of the drug in the same way that

Prozac permanently affected it after I discontinued its use many

years ago.

Whether this turns out to be anything like a magic bullet or not, It

definitely points to 5HT2 antagonism and perhaps, to a lesser degree,

5HT1 agonist activity rather than down-regulated dopamine pathways as

a likely cause for PSSD.

Will keep you posted.

What we've found is that increasing the sensitivity of 5ht2a receptors

leads to improved orgasm and improved wakefullness and also heightened

desire, and also brings back the sensitivity to the genitals.

Are you sure that this stuff is working?

i was reading a few posts from a month or 2 ago about nefazodone and

just wondered what happened to the guy that felt his pssd problems had

decreased as a result of taking it or if anyone else has tried it with

positive results. the reason i am very interested is that some years

back, before i ever knew about the existence of pssd i was prescribed

it due to its lack of sexual side effects. shortly after i started

taking it i became involved with a girl and remember that the few

months that i was with her sex was absolutely great. at the time i

thought that it felt so good simply because sex had been so bad for

some time, having been on another ssri for quite a while. now that i

look back, however, i actually think that the sex was even better than

normal. this makes me extremely curious about whether or not it could

help with pssd. back then i literally just stopped taking my previous

ssri and began taking nefazodone. i wonder if i hadn't started it

immediately if i would have developed pssd at that time as opposed to

last year after stopping nefazodone, due to its inefficiency at helping

with my anxiety and depression, and taking paxil for a few years. any

thoughts anyone?

I'm the guy who took nefazodone and still take very small maintenance

doses to counteract PSSD. 5HT2A blockade seems to be effective at

reversing some of the symptoms of PSSD, especially genital numbness

and anorgasmia.

One of the members here can't understand why an antagonist should

work; that only 5HT2 agonists should be helpful. Well, I'm not a

biochemist amateur or otherwise. I have no scientific credentials that

qualify me as an authority in this field, but I do know what works for

me and what doesn't. Nefazodone works in low doses 50mg/day or less.

Nefazodone antagonizes 5ht2a postsynapticly, so it might be a matter

of presynaptic vs postsynaptic as to whether an agonist or antagonist

will work appropriately. Perhaps, pssd24 can chime in.

It is unusual because we found that nutmeg and MDMA work very well

(particularly MDMA).

Does it work whilst taking it or whilst coming off?

Some thingsa which blockade 5ht2a receptors up-regulate them, but most

things that antagonise them just downregulate them.

Does it have any direct efect on nitric oxide or 5ht1a receptors, or A2

receptors etc?

How would you compare it with nutmeg?

OK I thought I ought to have a look at this drug.

It blocks the A1 androceptors as well as the 5ht2a recptors.

From what I've read, blocking androceptors can raise levels of several

chemicals including dopamine and not just adrenaline.

It probably has obscure effects upon sleep which I believe causes

changes in libido and nitric oxide levels, which researches don't

always look into (recovery sleep after sleep deprivation causes full

recovery from PSSD for up to 48 hours, it seems). Does this drugh make

you dream much less? Do you sleep more or less?

I genuinely don't think the 5ht2a antagonist would raise libido. If

anyhting it would lower the Nitric oxide levels, and also lower

prolactin, but also make it more dificult to get an erection

(experiments on rats have proven this).

I'm thinking of trying buspirone next.

I'm also curious as to how a 5ht2 antagonist can do this...

If it really works for anorgasmia and numbness that would be wonderful.

Anyone else had positive experience with nefazodone?

Main thing about nefazodone is that it is no longer on sale, which sucks.

Well that settles it. You shouldn't use nefazodone, but I will

continue using it in small doses. I'm not interested in theories. The

theory behind SSRI's sounded very promising at one time. Now I wish I

had never taken it.

Hvae you tried nutmeg yet? it saeems to be stronger than a lot of

meds that are supposed to be very strong.

There is some evidence that antagonizing 5-HT2a receptors might help

PSSD.For example, flibanserin, a drug in phase 3 trials for treating

sexual dysfunction, is a 5-HT2a antagonist (and 5HT1a agonist).

Also, ketanserin, which has been used off-label for years for

treating sexual dysfunction, is another 5-HT2a antagonist. Trazodone

is another 5HT2a antagonist that sometimes has the side effect of

priapism (not a good thing, but it's obviously, pro-erectile).

There is something else that I had forgotten. 5ht2a antagonists to

some extent block REM sleep, which can enhance the hypothalumus.

Experiments on the 5ht2a/5ht2b/5ht2c receptors have suggested that

5ht2a enhances libido, 5ht2c lowers testosterone, and 5htb does not

effect libido.

The 5ht receptors are usually unpredictable though. 5ht2a enhances NO

through cGMP, but also enhances other systems which reduce libido.

mCPP, buspar, and other serotonin agonists are not effective in

reversing PSSD, precisely because they overdrive the 5HT receptors in

the zona incerta which lies in the dorsal hypothalamus.

According to an abstract in Neuroendocrinology:

" Previous findings show that there is an inverse relationship between

serotonin (5-HT) activity in the ZI and plasma luteinizing hormone

(LH) levels, indicating that the 5-HT system in this area has an

inhibitory effect on LH release "

Luteinizing hormone triggers testosterone production and

spermatogenesis in the testes, which causes most of the effect known

as libido.

Nefazodone blocks 5HT2A in general, so it must act to attenuate

serotonin at 5HT2A receptor sites in the ZI. I have personally tried

buspar and it did absolutely nothing for libido. Per PSSD24's

suggestion, I have also tried mCPP...nothing but a baaaaad trip!

However, within 2 hours of resuming a 50mg tablet of Nefazodone, I was

able to have sex with my girlfriend

If you look up 5HT receptor on wikipedia it gives you a list of some

of the known antagonists for each receptor. Yohimbine HCL is a 5HT1A

antaogonist, so maybe I'll use a little of this along with my small

doses of nefazodone.

Nefazodone's antagonist action is the closest thing to a reversing

--have you tried trazodone (desyrel)? seems quite similar to nefazodone, but

without the liver risk..

i'm tempted to give nefazodone a try, but the liver issue is scary.

I was prescribed desyrel a long time ago, and found it only made me

extremely drowsy. I was nervous about using nefazodone, but given the

low incidence of severe liver damage (1 in 250-300,000 patient years)

and the fact that 68% of the cases were female, I felt that low doses

were safe to try (women might want to stay away from this drug)

I should point out that for me nefazodone loses effectiveness after

only two or three 50 mg doses at which point I take a couple of days

" drug holiday " and then use it again. I'm not sure why, but all of

these drugs monkey with feedback circuits in our brains.

Get a baseline liver panel test,and get a follow up 3-4 weeks later.

on paper they look quite similar (though i believe you that

nefazodone is better).

I recently talked with my psychiatrist, who actually cares about my

problems(unlike all other ones I've dealt with). I told him that I

was excited about people improving with serzone and the wikipedia

article saying that it is sometimes used as an antidote for pssd. He

said the hospital discontinued it because of the rare liver damage

liability that has everybody scared because it is fatal and that is

also why he is not allowed to write me a prescription. He also

didn't see any reserves anywhere in the computer system(also I live

in the US) He said that if I can get my hands on some(I think he was

hinting about the internet but I'm not sure:)), then he would check

my liver enzymes frequently so that if anything out of the ordinary

happens he can discontinue it immediately(there should only be

danger if continued after that unmonitered). I searched the internet

in my spare time the last couple of days but the couple of sites

I've found that didn't say serzone discontinued said " not available "

when I clicked on serzone(I've never been the best at finding things

on the internet). Can sssssssri or anyone else on the board point me

to a website where I can get it. I want to resort to trying to find

and pay another outside doctor and hospital and go through all the

motions as a last resort. Also thanks to sssssssri for posting and

Vornan for creating this site!

I have tried MDMA, it is the only drug that worked 100%. BUSPAR is a

mixed 5ht agonist (5ht1) and antagonist (5ht2). It is kmore likely to

lower libido than anyhting else. If you took too much McPP it would

not work - you are not supposed to get a bad trip from McPP. However,

what you have said is contrary to most expreiments with McPP which

show that it is an aphrodisiac.

You have made a big overgeneralisation about 5ht. 5ht has at least 15

distinct sites, all of which are different. Which particular 5ht

receptor is involved regulating LH?

A lot of people are feeling drowsy whilst taking that 5ht2 antagonist

stuff huh? Probably because 5ht controls the central nervous system

and sleep.

This antagonist would lower cGMP, meaning poorer smooth muscle

relaxation, and also lower dopamine release through 5ht2c receptors.

I had a quick look at the link. First of all, as far as I can see it

is primarily the 5ht1a receptors which control leutinizing hormone.

MCPP works on the 5htcc receptors. You seem to be suggesting that all

5ht agonists have similar properties? Not true, Buspar is a 5ht2

antagonist - completely different from MDMA or mcPP. I don't see why

you would compare the two drugs just because they are both serotonin

agonists; one serotonin site is usually completely different from

another in terms of the effects upon the central nervous system.

Serotonin has a very mixed and unpredictable effect upon libido, and

it depends upon which part of the brain the transmittors are, and

also which transmittors are being stimulated. The 5ht2c receptors

always enhance erections, but if you took too much mcPP, you would

produce too much 5ht, which would have negative effects.

Most experiments with 5ht2c agonists prove that they are very

important for erections. I can dig out more if you want.

The effect of Nefazodone is probably very quick, from what you have

suggested. I have had another look, and it may be the blockade of

5ht2c receptors which causes the effect. The fact that it also

antagonizes 5ht2c receptors would increase dopamine release.

I would have thoguht that it might lead to less strong erections,

through lack of cGMP, and 5ht2c antagonism, but maybe increased

orgasm due to increased dopamine levels.

Does it not also make you feel more sleepy in the morning?

The only thing I have tried that works 100% for PSSD is very long

periods of sleep deprivation, or MDMA (both of which increase 5ht2a

and 5ht2c binding a lot, and increase dopamine and nitric oxide).

But anyaway, what has your experience been with Nefazodone? If it

does work I would like to know in what way? does it increase the

strenght of erection or just orgasm?

Your doctor is partially correct. Serzone is not manufactured anymore

by Bristol-Meyers Squibb. But I am surprised that your doctor did not

inform you that nefazodone - the generic name - is still available

from TEVA USA who probably specialize in generic formulations.

I should mention some new observations.

1. Nefazodone acts not only as a 5HT2a antagonist it also acts as a

5HT2C agonist. I had reported earlier that nefazodone lost its

effectiveness after a couple of doses. Now I realize why. The 5HT2C

agonist action must actually dampen LH production in the hypothalamus,

resulting in lower testosterone overall.

I took 10 mg of generic fluoxetine (Prozac) with the 50 mg nefazodone

and got a sure fire increase in libido.

2. I don't think you have to take even 50mg nefazodone for our

purposes.Keep in mind we only want to antagonize 5HT2A. Nefazodone is

normally prescribed at doses starting at 100 mg and titrate up to

300mg, but that is if you're taking it as an atypical antidepressant

or anxylotic. In short: just a third to a quarter of one 100 mg tablet

might be enough as long as its taken with prozac which counteracts the

agonist action of Nefazodone at 5HT2C.

Amplidepressant. Please let me know what your doctor says about this.

I'd hate to find out that the generic version of serzone has also been

discontinued!

You said Buspar is a 5HT2 antagonist. It is not. It is a 5HT2 agonist.

So I certainly wasn't comparing Buspirone and mCPP because one is an

agonist and the other is an antagonist.

that lasted 8 hours, and that it had no effect whatsover on libido.

I compared nothing. I simply reported that my physiology did not

respond to the actions of these chemicals.

Ritanserin works at 5HT2 to prevent inhibitory effect of 5-HT and

8-OH-DPAT.

Ritanserin is a 5HT2 antagonist, so is nefazodone. But you bring up a

good point 5HT1 antagonism is also important. That's why I'm ordering

Yohimbine HCL (listed on wiki as a 5HT1 antagonist)

one of Nefazodone's metabolytes is mCPP so Nefazodone antagonizes

What is the net effect of Nefazodone ?(now combined with 10mg

fluoxetine to counteract the 5HT2C agonist effect which may well be

too powerful) I get a strong erection and ejaculate like normal. Does

it work reliably? not as much as I like, but I need to fine tune the

dosages for best results. I'm beginning to think less is more.

I've tried buspar which made me feel brain dead (i seem to recall it

also acts as a dopamine antagonist)I've tried the mCPP - poor man's

ecstasy - no good. Why would anyone want to rely on X for the rest of

their lives anyway? Its illegal, you don't know its purity, its

psychoactive, and it may have its own long term detrimental side effects.

Buspar is not known to be effective.

My point about MDMA is not that it is good to take it all of the

time, but merely that it appears to work, and that raises the issue

that it could be problems with serotonin and dysregulation that

causes pssd. This would lead to a signalling cascade and sleep

problmes which would effect smooth muscle relaxation, wakefullness,

and many other chemicals.

Its unlikely that people with pssd have really high serotonin. SSRI's

damage serotonin cells in the brain.

MDMA is toxic yes, but these other drugs cause liver poisoning.
